چکیده

Title of Document: EFFECTS OF TEMPERATURE ON GENE EXPRESSION AND SEX DETERMINATION IN THE MANGROVE RIVULUS, KRYPTOLEBIAS MARMORATUS Jennifer Strykowski, Master of Science 2011 Directed by: Assistant Professor Dr. Edward F. Orlando, Department of Animal and Avian Sciences Rivulus is a hermaphroditic, self-fertilizing fish species that possesses an ovotestis and, in the wild, exist as androdioecious populations comprised of hermaphrodites and males. Following embryonic incubation at an ambient temperature of 25 °C, rivulus develop normally as hermaphrodites. Embryos exposed to a lower temperature of 20 °C during a critical phase of embryogenesis develop as males. In this study, rivulus embryos were maintained at ambient (25 °C), low (20 °C), or high (31 °C) temperatures during various stages of embryogenesis. The expression of seven evolutionary conserved genes with known relevance to gonadal differentiation including figα, foxl2, cyp19a1b, cyp19a1a, dmrt1, sox9a, and sox9b was measured using real-time PCR. The expression of cyp19a1a was downregulated at 20 °C and the expression of ovarian-specific genes increased throughout embryogenesis. These results provide the first data documenting how temperature affects the expression of genes relevant to sex determination during embryogenesis in rivulus. The Genome of the Self-Fertilizing Mangrove Rivulus Fish, Kryptolebias marmoratus: A Model for Studying Phenotypic Plasticity and Adaptations to Extreme Environments

The mangrove rivulus (Kryptolebias marmoratus) is one of two preferentially self-fertilizing hermaphroditic vertebrates. This mode of reproduction makes mangrove rivulus an important model for evolutionary and biomedical studies because long periods of self-fertilization result in naturally homozygous genotypes that can produce isogenic lineages without significant limitations associated with i...

Population genetics and evolution of the mangrove rivulus Kryptolebias marmoratus, the world's only self-fertilizing hermaphroditic vertebrate.

The mangrove rivulus, Kryptolebias marmoratus (Rivulidae, Cyprinodontiformes), is phylogenetically embedded within a large clade of oviparous (egg laying) and otherwise mostly gonochoristic (separate sex) killifish species in the circumtropical suborder Aplocheiloidei. It is unique in its reproductive mode: K. marmoratus is essentially the world's only vertebrate species known to engage routine...

"Mangrove 'killifish': an exemplar of integrative biology": introduction to the symposium.

The mangrove rivulus, Kryptolebias marmoratus, (hereafter, rivulus) is one of the only two vertebrates known to self-fertilize, with the other being a recently named close relative, Kryptolebias hermaphroditus (Tatarenkov et al. 2012). Rivulus is also the first fish species found to have environmental sex determination, whereby lower temperatures inhibit ovarian development, thus providing one ...

Complete mitochondrial genome of a self-fertilizing fish Kryptolebias marmoratus (Cyprinodontiformes, Rivulidae) from Florida.

The complete mitochondrial genome was sequenced in a mangrove rivulus Kryptolebias marmoratus from western Florida using next-generation sequencing. The 17 329 bp-long genome was identical in length and 99.8% similar to a previously published genome of this species from a specimen of unknown geographic origin. Gene arrangement in K. marmoratus is similar to other cyprinodontiform fishes, except...

Phenotypic plasticity and integration in the mangrove rivulus (Kryptolebias marmoratus): a prospectus.

The mangrove rivulus (Kryptolebias marmoratus) is a small fish native to mangrove ecosystems in Florida, the Caribbean, Central America, and South America. This species is one of only two self-fertilizing, hermaphroditic vertebrates capable of producing offspring that are genetically identical to both the parent and all siblings. Long bouts of selfing result in individuals with completely homoz...
